Q: Will this work with my D3000?

It should work with a D3000, D3200, etc, but only as a manual focus lens. Some of Nikon's lower-end DX (cropped sensor, APS-C format) have electronic contact points to work with lenses that have their own built-in AF motor, but do not themselves have a motor to move the lens's AF. Those cameras will only auto focus with AF-S or AF-I lenses, but not AF-P lenses. If you are using this lens on one of these cameras and the shutter release button won't take a picture, the camera may be preventing shutter release because you don't have a clear focus at your selected AF point. (Perhaps you focused on one object and then reframed the shot.) You could check to see if you have a menu setting for "AF-S priority selection" that might allow the camera to shoot when it thinks it is out of focus. Otherwise, even though you are focusing manually, you would probably need to lock focus and then reframe so the camera knows that you intend that focus.

Q: Will this work on a D5500 without a firmware update?

The number of cameras compatible with AF-P NIKKOR lenses is limited. Even for compatible cameras, firmware update may be required*. Fully compatible models: D850, D500, D7500, D5600, D5500, D5300*, D3400, D3300* and later models Compatible models with limited functions: D5, D810 series, Df, D750, D7200, D7100, D5200, Nikon 1 series with the FT-1 Incompatible models: D4 series, D3 series, D2 series, D1 series, D800 series, D700, D610, D600, D300 series, D200, D100, D7000, D5100, D5000, D90, D80, D70 series, D3200, D3100, D3000, D60, D50, D40 series, film cameras... So, in short, YES it will!
