Q: It states for hot tubs to remove the filter however my understanding is a hot tub should not be run without a filter. Will it damage the filter?

I can speak for your hot tub, but ours can be run for a short time with it, I always remove my filter to clean that separately, soaking it over night.

Q: I can't imagine that 50-75ml of this product can replace true spa flush, which normally requires about 250ml or more to rid a hot tub of biofilm?

Hi, I can't help too much, but that is what it says on the container. It says you should do this every 2 -3 weeks though. For your information It does recommend 250ml for hot tubs as well .

Q: How would I use this to clean a Miami layz hot tub? Does it need to be empty or full?

Not sure my jacuzzi bath fill to cover top jets run for 15 minutes then rinse to clean
