Will this have cross platform with WiiU?

This hasn't been confirmed, but it's highly unlikely as this version has new modes and pokemon that aren't in the WiiU Version. I think if it had cross play it would have been announced by now.

Does it come the Shadow Mewtwo amiibo card?

No it doesn't however you'll be able to play as Shadow Mewtwo in the game.
