ALL Electrolytic capacitors should have polarized marks. These are marked plus and minus. If you need non polarized the you should look for different capacitors. If you need them for say, speaker crossover networks, you need to order Bipolar electrolytic . All general purpose electrolytic are polarized and polarization must be observed. Voltage rating must be equal to or greater than the voltage expected in the circuit.

Can I substitute a 4.7 uf @ 16v with a 4.7 uf @ 400v ?

Yes, you can always use a higher voltage with same C value.
