One nibble is enough to do for a rat but if all your blocks have gone then that is the time to replace them. The difficulty is that a large infestation will need more blocks to deal with it. After a short time, the blocks will remain untouched. After that, check them occasionally in case there is a new infestation. **Q: Is it harmful to squirrels?** A: Yes it's harmful to anything that eats it. That's why it needs to go into the rat holes not left where they pass because others animals will find it.
