Q: Will you need to put soil in the baskets?

These planters are for aquaponics/hydroponics. So you need to use a grow media that isn’t soil like expanded clay pebbles or rock wool cubes etc. Soil would more than likely wash out.

Q: Will standard 2" mesh pots work as a replacement

No, standard net pots would not work if they are flat on the bottom. If you look at the pictures, these are tapered at the ends to lock into the float.
